Dans son livre Directs du droit, Eric Dupond-Moretti écrit être devenu avocat par « détestation de la peine de mort ».

En 2008, il préside le comité de soutien de la socialiste Martine Aubry pour les élections municipales à Lille.

A partir de juin 2012, il est l’avocat d’Abdelkader Merah, accusé de complicité dans les attentats perpétrés par son frère Mohamed Merah et condamné en avril 2019 pour « association de malfaiteurs » et « complicité d’assassinats ».
En avril 2015, il est l’avocat de David Roquet poursuivi pour proxénétisme aggravé et escroquerie en lien avec l’affaire DSK et déclare que ce sont « des copains qui s’offrent du bon temps ».
En mai 2015, il déclare qu’il faut « interdire le Front National ».
En 2016, il est l’avocat de Karim Benzema mis en examen pour sollicitation de prostitué mineure » et obtient sa relaxe. La même année, à la demande du roi du Maroc Mohamed VI, il assure la défense du chanteur marocain Saad Lamjarred accusé d’agression sexuelle et de viol.
En février 2018, il est l’avocat de Jérôme Cahuzac, ancien ministre accusé de fraude fiscale.
En avril 2018, il est l’avocat de l’ancien Secrétaire d’Etat Georges Tron accusé de viols en réunion sur plusieurs de ses collaboratrices et obtient son acquittement.
En 2019, il est l’avocat de Patrick et Isabelle Balkany
